The Nonenzymatic Preparation in Solution of N- (S-Phosphoribosyl) anthranilic Acid, an Intermediate in Tryptophan Biosynthesis*

چکیده

Yanofsky (1) originally proposed N-(5’-phosphoribosyl)anthranilic acid as the first intermediate after anthranilic acid in tryptophan biosynthesis in microorganisms. Evidence was subsequently found which suggested that this compound is formed enzymatically from anthranilic acid and ribosylpyrophosphate 5-phosphate (2, 3). Although the dephosphorylated form of PRA,’ N-ribosylanthranilic acid, has been synthesized and characterized (4), repeated attempts to synthesize PRA have been unsuccessful (2,5, 6). The elusiveness of this intermediate has been ascribed to its lability. For example, tryptophanrequiring mutants of microorganisms which would be expected to accumulate PRA or its dephosphorylated form are found to accumulate primarily anthranilic acid, which is presumably a breakdown product. Synthetic N-ribosylanthranilic acid has a half-lie of approximately 30 min at pH 7.0 and 37” (5). It is the purpose of this communication to describe an extremely simple method of preparing PRA in solution which makes it available as a substrate for enzymatic studies.

In vivo regulation of intermediate reactions in the pathway of tryptophan biosynthesis in Neurospora crassa.

The in vivo regulation of intermediate reactions in the pathway of tryptophan synthesis in Neurospora crassa was examined in a double mutant (tr-2, tr-3) which lacks the functions of the first and last enzymes in the pathway from chorismic acid to tryptophan. The double mutant can convert anthranilic acid to indole and indole-3-glycerol, and the production of these indolyl compounds by germinat...

The Conversion of Shikimic Acid to Anthranilic Acid by Extracts of Neurospora Crassa.

The role of shikimic acid as a common intermediate in the synthesis of phenylalanine, tyrosine, tryptophan, and p-aminobenzoic acid has been recognized for some time (l-3). The enzymes which catalyze the synthesis of shikimic acid from intermediates of carbohydrate metabolism have been observed in extracts of bacteria (4-6) as well as Neurospora crassa (7). On the other hand, the reactions lead...

Enzymatic Formation of Catechol from Anthranilic Acid.

Anthranilic acid is known to be formed from kynurenine in animals and microorganisms (3-6) and to be degraded by cell-free extracts of a Pseudomonas species adapted to L-tryptophan (7). Catechol has been postulated as an intermediate in the catabolism of anthranilic acid on the basis of experiments utilizing sequential enzyme induction (8). Several groups of investigators have recently succeede...

Multi-walled Carbon Nanotubes/Ionic Liquid Nanocomposite Modified Carbon-ceramic Electrode: Electrochemistry and Measurement of Tryptophan in the Presence of Uric Acid

In the present work, an efficient modified carbon-ceramic electrode by multi-walled carbon nanotubes/ionic liquid nanocomposite (MWCNTs/IL/CCE) was prepared through a simple and repeatable procedure. The introduced modified electrode was used for the study of the electrochemical behaviour and determination of tryptophan (Trp) in the presence of uric acid (UA) using cyclic voltammetry (CV), diff...
